Problem
For your lab experiment, you received a solution already made containing both 16S rRNA primers, 16SF08 and 16SR08, for amplifying by PCR a portion of the 16S rRNA gene from DNA you prepared from bacterial cells living in mosquito gut. The concentration of each primer was 20 ng/µl. This solution was prepared from the primers stock solutions at the following original concentrations:
Primer 16SF08 stock solution: 1.24 µg/µl
Primer 16SR08 stock solution: 1.16 µg/µl
Write below the volumes in µl of each stock solution as well as the water volume in order to make 200 µl of the primers solution at the concentration of 20 ηg/µl each primer, the two primers in one solution. Round the volumes values to one decimal.
1. Show the calculations of the primer stock solutions volumes and write down the numbers in the table below:
Stock solution Volume (in µl)
1.24 µg/µl 16SF08 _______
1.16 µg/µl 16SR08 _______
Water _______
Total _______
2. Write down your line of thinking to calculate the above volumes.
