Isolation of dna fragments bound strongly with protein
1. Through pcr
Pcr permits isolation of dna fragments from genomic dna through selective amplification of a specific area of dna. Large amount of pure dna fragments bound by protiens can be seperated.
2. Chromatin immuno precipitation
This technique gives one way of empirically finding the sites of dna that a known gene regulatory protein occupies under specific set of conditions. In this technique proteins are covalently attached to dna to be isolated, dna is mechanically sheared into minute fragments. Antibody directed against a known regulatory protein is then utilized to purify dna which is covalently attached to the protein. This dna fragments are hybridized to micro arrays that comprises of the complete genome displayed a series of dna fragments, the precised genomic location of every dna fragment can be found.
