Problem
Your colleague cut linear DNA:
with Ecor-1 and obtained 1 band on the gel, 2200bp
with BamH1 and obtained 600 and 1600
with PvuII and obtained 400 and 1800
with BamH1 + PvuII + EcorI 200bp, 400bp, 1600
Control: linear DNA without any enzyme: 2200bp
Provide a table with the number of bands and the number of cuts with each individual enzyme for the linear DNA, be sure an include the co-digest.
Draw the individual restriction maps for each digest of the linear DNA. Be sure and label enzymes and fragment sizes on the map. Next provide the composite map with all enzymes. Be sure and identify the enzyme name and the size of the fragment on all the maps.
Now assume this is a plasmid, and you keep the number of cuts the same as you have with the linear DNA, provide many bands would you expect to with each enzyme and the co-digest with plasmid DNA?
