Question: You have a recipient strain that is auxotrophic for cysteine, histidine, and alanine and a HFR donor strain that is prototrophic for these three markers. The recipient strain is resistant to the antibiotic, kanamycin while the donor strain is not. You carry out conjugation and then plate the cells on media as shown below. Kanamycin is added to the media to kill off the donor strain after conjugation was allowed to occur. Only the recipient strain should grow on the plates.
