Scanning Electron Microscope (SEM):
Scanning Electron Microscope is employed to inspect the surfaces of microorganisms. Scanning Electron Microscope gives a 3-dimensional view of the specimen. The electron gun generates a finely focused beam of electrons termed as the primary electron beam. Such electrons pass via electromagnetic lenses and are directed over the surface of the specimen (figure is as shown below).

The prime electron beam blocks electrons on the surface of specimen, and the secondary electrons therefore generated are transmitted to an electron collector, amplified and utilized to generate image on a viewing screen or photographic plate. The image is termed as a Scanning Electron Micrograph.
