Phenol-coefficient technique is a tube-dilution method universally followed official test.  Phenol-coefficient is stated as the ratio among the greatest dilution of the disinfectant killing the test organism in 10 min and not in 5 min to the highest dilution of phenol killing the test organism in 10 min and not in 5 min. Since phenol is employed as standard for comparison this is termed as phenol-coefficient technique and is appropriate for testing disinfectants miscible with water and contain antimicrobial action identical to phenol. A series of dilution of the disinfectant tested is made and dispensed in 5ml amounts in test tubes to which 0.5ml of 24h old broth culture of test organism either Salmonella typhi or Staphylococcus aureus is inoculated. Concurrently a sequence of dilutions of phenol is made and the similar quantity of inoculum is added. Both the set of tubes, those having the disinfectant and the test organisms and phenol and the test organisms are located in a water bath at 20oC.  Subcultures are prepared with the inoculation needle into sterile tubes of medium at 5, 10 and 15 min intervals, incubated and noticed for growth. The highest dilution of the disinfectant tested killing the test organism in 10 min and not in 5 min is sub-divided by the highest dilution of phenol presenting similar outcome to determine the phenol coefficient. An illustration is given in Table & figure shown above.
