Laboratory Diagnosis:

Selection of Specimen:

1. Pus is gathered with granules.

2. Swabs from deeper portions of the infection.

3. The scrapings of the deeper portions [i.e., superficial parts might be contaminated with bacteria]

Transport:

Specimen must be placed in a sterile vessel, promptly transported to the laboratory to avoid contamination. The granules must be washed with sterile saline repeatedly to eliminate the bacteria. Color, consistency, size, microscopical appearance of granules must be observed.

Granules are crushed and inspected in saline and KOH.

Gram stain:

The granules are compressed, fixed and Gram stained. The filaments are Gram positive. From the size of the filament it is recognized whether it is Eumycotic or Actinomycotic granules.
