Dark-field Microscope:
The dark-field Microscope is employed for examining live microorganisms that are either invisible in the normal light microscope, can’t be stained by standard techniques or are so distorted by staining that their characteristics can’t be recognized.
The condenser in dark field microscope has an opaque disc. The disc blocks light which enters the objective lens directly. The light which is reflected off the specimen enters the objective lens. As there is no direct background of light, the specimen emerges light against a dark background, therefore the name dark-field Microscope (figure is as shown below).
