There are two main models that explain the formation of the enzyme-substrate complex: the lock and key model and the induced fit model. In lock and key model enzyme has a region with specific spatial conformation for binding of substrate. In induced fit model binding of substrate involves a change in spatial configuration of enzyme for substrate to fit.
